Aceto-Orcein Stain

 

Ingredients
orcein
1 gm
 
glacial acetic acid
50.0 ml
 
85% lactic acid
50.0 ml
 

 

Directions

Warm gently (do not boil),
then cool and filter.

Used to stain chromosomes in chromosome squashes of Drosophila salivary glands. Stain for 2-5 min. prior to making the squash.

 

Mayer’s Carmalum

 

Ingredients
Stock solution:
carmine
1 gm
 

ammonium alum (aluminum ammonium
sulfate)

10 gm
 
distilled water
200 ml
 
Boil 20 minutes.
Working solution:
carmalum stock solution
5 ml
 
glacial acetic acid
0.4 ml
 
distilled water
100 ml
 

 

Directions

Mix ingredients for working solution. When dissolved, filter.

Add 1 ml formalin to inhibit mold growth. Will keep indefinitely.

Stains nuclei. Is a progressive stain. Specimens are usually adequately stained in 4-8 hours. Stop staining when specimen is a deep pink-red color.
